Profinity eXact融合標(biāo)簽系統(tǒng)提供兩種pPAL表達(dá)載體試劑盒。兩種試劑盒都包括含5901個(gè)堿基的pPAL7載體，一種是含有通用的、預(yù)先酶切的載體，可插入任何目標(biāo)基因，而無論內(nèi)切酶位點(diǎn)如何;另一種是超螺旋的質(zhì)粒。pPAL載體由基于T7的表達(dá)質(zhì)粒衍生得到，應(yīng)用了T7 lac啟動(dòng)子和終止子，以及T7 RNA聚合酶表達(dá)宿主，用于誘導(dǎo)目標(biāo)蛋白的表達(dá)。該質(zhì)粒具有氨芐青霉素( ampillin)的抗性，lac1表達(dá)抑制以緊密控制目標(biāo)基因的轉(zhuǎn)錄，同時(shí)還有衍生自pMB1的ColE1的復(fù)制起始點(diǎn)。目標(biāo)基因克隆到Profiniy eXact pPAL載體中后表達(dá)了作為融合序列(或標(biāo)簽)的枯草桿菌蛋白酶的N端前結(jié)構(gòu)域( Prodomain)。

pPAL7不依賴于限制性內(nèi)切酶克隆( RIC，restriction- independent cloning )的即用型表達(dá)載體預(yù)先以Sap1和EcoR1對(duì)超螺旋質(zhì)粒進(jìn)行雙酶切，然后進(jìn)行去磷酸化處理以減少連接后背景轉(zhuǎn)化株。

pPAL7超螺旋質(zhì)粒表達(dá)載體用于常規(guī)的目標(biāo)DNA的克隆，采用傳統(tǒng)的限制性酶切克隆策略。

BL21 ( DE3) E..Coli細(xì)胞*是基于T7載體的蛋白質(zhì)表達(dá)的**宿主。該細(xì)胞是DE3λ溶原性細(xì)菌，在lacUV5啟動(dòng)子控制下帶有T7 RNA聚合酶基因。T7 RNA聚合酶產(chǎn)物由IPTG誘導(dǎo)，該誘導(dǎo)引導(dǎo)了載體中定位于T7 lac啟動(dòng)子下游的目標(biāo)基因的表達(dá)。該菌株具ompT和lon蛋白酶缺陷，這提供了改良的重組蛋白的穩(wěn)定性。
